Inhibition of cAMP-induced CGRP receptor internalization. CLR-FAP tagged CGRP receptor internalization was measured using flow cytometry and graphed as percent of control mean fluorescence intensity (POC). Increasing CGRP concentrations resulted in CLR-FAP-tagged CGRP receptor internalization (open triangles) with a correlative decrease of membrane-bound CLR-FAP-tagged CGRP receptors (closed triangles) (a). In the presence of 100 nM CGRP, AA58 (b) or 8E11 (c) inhibited internalization (open squares) and concomitantly increased membrane-bound CLR-FAP-tagged CGRP receptors (closed squares), whereas no intrinsic effect was observed in the absence of CGRP (open and closed circles)
